A global metabolomics minefield: Confounding effects of preanalytical factors when studying rare disorders

A common challenge when studying rare diseases or medical conditions is the limited number of patients, usually resulting in long inclusion periods as well unequal sampling and storage conditions. The main purpose this study was to demonstrate challenges comparing samples subject different preanalytical We performed a global (commonly referred “untargeted”) liquid chromatography-high resolution mass spectrometry metabolomics analysis blood from cases sudden infant death syndrome controls stored dried spots on chemical-free filter card for 15 years at room temperature compared with same whole −80°C before preparing new using chemically treated card. Principal component plots distinctly separated based type storage, but not versus controls. This emphasizes importance thoroughly addressing factors during design interpretation, enabling identification real, biological differences between sample groups whilst preventing other random variation be falsely interpreted results.
